Figure. (A) PC5-asODNs-1 and -2 reduced 2.5%FCS-induced PC5 protein levels (117 kDa) in a concentration-dependent manner, whereas PC5-asODNs-3 had no effect. α1-collagen targeting asODNs were used as control.21 co indicates serum-deprived VSMCs; TM, transfection medium. (B) Representative in vivo findings of VSMC transfected with a FITC-labeled PC5-asODNs-2. Without the use of liposomal transfection medium, PC5-asODNs-2 is found only in the cytoplasm (A), whereas with transfection medium, asODNs enter the nucleus (B). (C) PC5-asODNs-2 (1 μM) specifically inhibited 2.5%FCS-induced PC5 protein levels, while sense (se) and scrambled (scr) sequences had no effect. Levels of furin (87 kDa) were not affected by PC5-asODNs. Reblotting with α-SMA was done to demonstrate equal protein loading. Densitometry of PC5 inhibition by asODNs (B) is depicted in (D) (#P<0.05 vs. controls).
