(Microsoft PowerPoint) (73.0 kb). Kinetic Analyses of Heterologously Expressed HERG Variants. Each variant and WT-HERG were expressed transiently in CHO cells using IRES2-EGFP plasmid constructions. The voltage protocol employed for each experiment is shown in the inset (arrow indicates the time point at which currents were compared between variants: for D, the time constants were determined over the 3 second repolarization interval). (A) No significant differences were observed among the three alleles in the time constants of inactivation (N = 8-10). (B) There were no significant changes in the voltage-dependence of inactivation (N = 6-9) among the three constructs. (C) Time constants for recovery from inactivation (N = 9-11) were larger at –40 to –20 mV for A1116V as compared to WT-HERG, while there was no difference between K897T and WT-HERG. (D) There were no significant differences in either the fast or slow deactivation time constants (N = 7-9) among the three alleles.
